水土中总磷酸盐可见分光光度法检测试剂盒测定步骤:
1.加样
1. 除包被外都需45度加样
2.加样体积要准确
3.管底加样,不能加在管壁上
4.加样时不能产生气泡
2.温浴
1.加标本后和加结合物后,应立即放入按规定的反应温 度的水浴箱。
2.各ELISA板不应叠在一起。
3.为避免蒸发,板上应加盖,或将板平放在底部垫有湿 纱布的金属湿盒中。
4.加入底物后,反应的时间和温度通常不做严格要求。 如室温高于20℃,ELISA板可避光放在实验台上,以便 不时观察,待对照管显色适当时,即可终止酶反应。
3.洗涤
1.洗涤在ELISA过程中不是反应步骤,但却 是决定实验成败的关键。
2.目的是洗去反应液中没有与固相抗原或 抗体结合的物质以及在反应过程中非特 异性吸附于固相载体的干扰物质。
4.读板
1.阴性对照颜色极浅,在定性测定中一般 可采用目视比色。
2.如用酶标仪测定结果,准确性决定于 ELISA板底的平整与透明度、酶标仪的质 量和软件的算法。
